Autoimmune orbitopathy has been reported in patients with autoimmune thyroid disease. The mechanism of this association is unknown. We sought to determine whether the prevalence of orbitopathy is higher in patients with Graves’ disease than in the general population and to correlate the prevalences of orbitopathy in our patients with thyroid autoimmunity, the presence of antibodies to thyrotropin receptor (TRAb), clinical features of Graves’ disease, or thyroid status. Fetal pituitaries undergo extensive growth in early gestation. Growth factor responses in pituitary stem/progenitor cells and their immediate descendants have not been examined. In this study, we compared the expression of epidermal growth factor receptor (EGFR) and its ligands in fetal and adult pituitaries, with a focus on the corticotroph lineage. Ectopic expression of EGFR in corticotroph-derived AtT-20 cells induces a mitogenic response to epidermal growth factor (EGF) and transforming growth factor (TGF)alpha. The cytoplasmic domain of EGFR is critical for the mitogenic response, because EGFR deletion mutants lacking the cytoplasmic tail of EGFR in AtT-20 cells fail to respond to TGFalpha. EGF receptor gene expression is highest in intermediate and basal lobe corticotrophs and lowest in anterior lobe corticotrophs. In situ hybridization revealed EGFR expression in intermediate and basal lobe corticotrophs, and the latter also expressed EGF and TGFalpha. Expression of EGFR family ligands, EGF and TGFalpha, has been reported in the developing pituitary gland, but these ligands are generally considered to be locally acting trophic agents in the developing pituitary gland.
